HPLC-ELSD法同时测定熟地黄中果糖、葡萄糖含量

Determination of fructose and glucose in Radix Rehmanniae Preparata by HPLC-ELSD

目的建立高效液相色谱-蒸发光散射检测法同时测定熟地黄饮片中葡萄糖、果糖的含量。方法色谱柱:ZorbaxRX-SIL(4.6×200mm);流动相:乙腈-水(95∶5);流速:1.0mL.min-1;柱温:25℃;ELSD:漂移管温度105℃,载气流速2.5mL.min-1。结果果糖在1.00～11.00μg范围内(r=0.99996),葡萄糖在0.95～10.45μg范围内(r=0.99991)线性良好,平均回收率分别为97.93%、98.65%,RSD分别为2.07%、2.48%(n=5)。结论该方法操作简便,可用来控制熟地黄饮片的质量。

OBJECTIVE To develop an HPLC--ELSD method for determination of fructose and glucose in Radix Rehmanniae Preparata. METHODS Chromatographic column： Zorbax RX- SIL（ 4. 6× 200mm） ; Mobile phase： acetonitrile--water （95 ： 5） ; flow rate： 1.0mL · min^-1. Column temperature： 25℃ ; ELSD： temperature of drift tube, 105℃ ; gas flow, 2.5mL · min^-1. RESULTS The assay displayed good linearity,the linear range of fructose was 1.00-11.00μg（ r =0. 99996）and that of glucose was 0.95-10. 45μg（ r =0. 99991）. The average recovery（ n =5） was 97.93%（ RSD =2.07%） and 98.65%（ RSD =2.48%）. CONCLUSION The method is simple and can be used for controlling the quility of Radix Rehmanniae Preparata
