摘要
目的:探讨骨髓造血细胞(hematopoietic cells,HCs)移植于克罗恩病(Crohn’s disease,CD)大鼠模型后其在大肠的定位情况。方法:用三硝基苯磺酸(TNBS)灌肠法制作SD大鼠CD模型。用悬浮法培养同种异体HCs,并用5-溴-2-脱氧尿嘧啶(BrdU)标记,经尾静脉注入体内,并以生理盐水为对照。于移植后第7天、第14天及第21天处死大鼠,取全段大肠组织,用免疫组化方法检测HCs的定位情况。结果:免疫组化显示移植组各时间段大肠组织均有BrdU阳性细胞定位,主要在黏膜上皮层,对照组阴性。结论:HCs可定位于大鼠CD模型的肠道中。
Objective:To investigate the localization of allogeneic bone marrow hematopoietic cells (HCs) in the colon of allograft murine models of Crohn's disease (CD). Methods: The HCs from allogeneic SD rats were cultureexpanded in vitro and labeled with BrdU before transplantation, and then transplanted into a TNBS enema induced allografted murine model of CD by tail vein injection. Normal saline was used as control. The BrdU immunoreactive cells were detected in the colon of the models by immunohistochemical staining at 7, 14 and 21 days after transplantation, Results: The BrdU immunoreactive cells were present in the colon of the allografted models at 7, 14 and 21 days after transplantation. Most of them were located in the mucous epithelium layer. No BrdU immunoreactive cell was found in the colon of controls. Conclusion : HCs can be localized in the colon of TNBS-induced murine models of CD, which may contribute to the regenerative repair of impaired colon tissues.
