摘要
目的:探讨维生素B6对糖尿病肾病(DN)大鼠肾功能的保护作用及可能的机制。方法:利用链脲菌素(STZ)腹腔注射法诱导建立DN大鼠模型,将其随机分为模型组(D组)、维生素B6治疗组(B组)、氨基胍治疗组(A组),并与正常对照组(C组)比较,每组10只大鼠;A、B组每天分别用氨基胍、维生素B61g溶于1L蒸馏水中,C、D组仅给予1L蒸馏水,24 h后自由饮用。观察治疗期间大鼠的一般状况、空腹血糖(FBG)、糖化血红蛋白(HbA1c)、血三酰甘油(TG)、血总胆固醇(TC)、血肌酐(SCr)、肾小球滤过率(GFR)、24 h尿白蛋白排泄率(UAER)、肾小球提取物糖基化终末产物(GTE-AGEs)、肾组织病理等改变。结果:(1)造模组大鼠均出现肾脏功能损害。(2)维生素B6对FBG及HbA1c无影响,但能降低DN大鼠的TG、TC、SCr、24h-UAER,增加GFR。(3)维生素B6能降低DN大鼠肾小球AGEs,其作用效果与氨基胍相当。结论:(1)维生素B6无降糖作用,但对肾脏功能有一定的保护作用;(2)维生素B6能降低糖尿病大鼠的高脂血症;(3)维生素B6具有与氨基胍相似的抑制肾脏AGEs在糖尿病大鼠肾脏蓄积的作用,发挥其对糖尿病肾病的防治作用。
Objective:To explore the effect of Vitamin B6 on renal function of streptozotocin (STZ)-induced diabetic nephropathy (DN)rats and its mechanism. Methods: Diabetic nephropathy rats induced by intraperitoneal injection of STZ were randomized into PN model group (group D, n = 10 ), Vitamin B6 group (group B, n = 10 )and aminoguanidine group( group A, n = 10 ), contrasted to anormal control group ( group C, n = 10 ). Aminoguqnidine and VitB6 with 1 g dissolved in 1 L distilled water were given to group A and B,while group C and D only had 1 L disstided water. All groups drunk water freely after 24 hours. During and after the treatment, the changes in general condition, fasting blood glucose (FBG) levels, hemoglobin A1 c (HbA1 c ) , blood triglyceride (TG) , total blood cholesterol (TC) , serum creatinine(SCr), 24-hour urinary microalbumin excretion rate (UAER), clearance of creatinine glomerular filtration rate (GFR), glomerular tissue extracts AGEs (GTE-AGEs) and renal tissue morphology were observed in different groups. Results:Diabetes mellitus and renal function impairment occurred in the model groups. Vitamin B6 had no effects on FBG and HbA1c levels, while it decreased TG, TC, SCr and 24 h UAER and increased GFR. Vitamin B6 also shown similar effects of aminoguanidine on GTE-AGE reduction. Conclusion : Vitamin B6 has no effect in decreasing blood sugar level, but has a definite effect in renoprotection, and decreases hyperlipidemia in diabetic rats. Vitamin B6 showed similar effects with aminoguanidine on parameters measured, which supports a mechanism of involving AGEs inhibition.
