白血病干细胞生物学特性的研究

摘要 目的 通过对白血病原代细胞的体外培养,研究白血病干细胞(LSC)的生物学特性.方法 分离18例白血病患者骨髓或外周血中单个核细胞(MNC),在体外进行培养观察,并用流式细胞术方法检测分离的MNC中LSC的数量及和细胞表面标记.结果 从白血病患者中分离的MNC在体外培养1周后基本死亡.流式检测分离的MNC中免疫表型是CD34^+CD38^-CD123^+的细胞群约有0.676%(0.03%~2%).结论 白血病干细胞难以在体外培养生长及扩增.外周血及骨髓中分离白血病患者的单个核细胞中,极少部分细胞表达干细胞标志. Objective To study the biologic characteristics of leukemia stem cells(LSC) by primary leukemia cells culture in vitro. Methods Mononuclear cells (MNC) of bone marrow or peripheral blood from eighteen leukemia patients were isolated and cultured in vitro. Cell surface marker was analyzed with flow cytometry. Results Leukemia primary cells from leukemia patients almost all died after one week's culture in vitro. CD34^+ CD38^- CD123^+ LSC was about 0. 676% (0.03% ~ 2% ). Conclusion Leukemia stem cell is hard to be cultured and proliferated successfully in vitro. Most mononuclear cells from bone marrow or peripheral blood dose not express stem cell markers.
