摘要
以表现柑桔裂皮病典型症状的罗伯逊脐橙为材料,采用双向电泳、RT-PCR和RNA斑点杂交及组织印迹杂交的方法,对柑桔裂皮类病毒进行了检测,结果表明RT-PCR和2种核酸杂交技术均可用于该类病毒的有效检测。对该类病毒全长RNA的RT-PCR产物进行了克隆和序列分析,该分离株(命名为CEVd-HB分离株)的全长RNA由371个核苷酸组成,与已报道的其它不同来源分离株相似性为90%~99%。
Citrus exocortis viroid (CEVd) in Citrus sinensis Osbeck cv. Robertson, which showed typical symptoms of CEVd infection, were detected by bi-directional PAGE electrophoresis, RT-PCR, RNA dot-blotting and tissue-printing hybridization. Results showed that RT-PCR and two hybridization methods were effective for the detection of the viroid. The RT-PCR products from the full-length genomic RNA of the viroid(named as CEVd-HB) were cloned. Results of sequence analysis indicated that the full-length genomic RNA of the viroid consist of 371 nucleotide acids and had similarities ranging from 90% to 99% with reported isolates of the viroid.
