摘要
目的研究神经干细胞移植对戊四氮慢性癫痫模型大鼠海马区r-氨基丁酸(r-GABA)和谷氨酸(Glu)及脑电图的变化。方法56只雄性大鼠选择雄性SD大鼠56只,腹腔注射生理盐水组8只,癫痫模型组分神经干细胞移植实验组24只,未移植对照组24只。然后3组中再分A,B,C,D4组。其中注射组生理盐水每组2只,实验组和对照组各6只。建立脑电电极。制作成四氮慢性癫痫模型,记录脑电图和高效液相色谱仪测定大脑海马组织GABA、Glu含量。结果移植组1周,2周GABA、Glu的变化不大,而到第4周后,GABA逐步升高,Glu逐步下降,第8周时GABA明显升高,Glu明显下降。未移植组GABA、Glu含量基本无变化,移植组对癫痫大鼠的脑电具有明显的抑制作用,移植组与未移植组比较P<0.05,组间有明显差异。结论神经干细胞移植可以干预癫痫大鼠海马区GABA、Glu的变化。
Objective To investigate changes of GABA and glutamate by neural stem cells transplantation in the hippocarapus of pentetrazole induced chronic klnding rats. Methods A model of chronic epilepsy in rats was established by pentylenetetrazol-kindled seizures. After neural stem cells transplantation, determined r-aminohutyric acid (GAB/t), glutamate (GLU) in hippocampus by means of high performance liquid chromatography. Results The level of GABA was significantly higher in hippoeampus by neural stem cells transplantation after 2 weeks, the lever in 8 weeks was higher than 2 weeks. Transplantation than that in normal control group respectively. The level of Glu was markedly lower. The level of GAB and Glu in normal control group keeps unsignificantly, there was significantly difference between transplantations and normal control groups, P〈0.05. Neural stem cells transplantation can decrease the frequency of discharge of the epileptic rats and reduce the amplitude of discharge to half its previous level.
