口服5-氨基酮戊酸在小鼠肝脏、胰腺中的代谢

摘要 目的:观察光敏剂5-氨基酮戊酸(ALA)口服后在肝和胰腺中的代谢,以获得理想的ALA剂量、荧光诊断及治疗时间。方法:给昆明小鼠口服不同浓度的ALA(20mg/kg、40mg/kg、60mg/kg、80mg/kg 4个组别)后,在不同时间点(1小时、2小时、4小时、6小时、8小时、10小时、12小时、24小时、48小时),以波长410nm半导体激光诱导荧光,并通过OMA荧光光谱仪分析检测组织荧光强度,了解ALA不同时间在肝脏、胰腺中的代谢状态。结果:荧光强度在口服ALA40mg/kg组后1小时最强。结论:口服ALA40mg/kg为最适诊断剂量,最佳检测时间为口服ALA40mg/kg后1小时。 Objective: To observe the metabolize status of oral ALA in liver and pancreas of mice and make up suitable dose of ALA in diagnosis and treatment time. Methods: Different dose of ALA was taked orally(20mg/kg、40mg/kg、60mg/kg、80mg/kg) , at different time point , tissue fluorescence was irradiated by semi-conductor laser(410nm) and fluorescence power was determined by optical mutichannel analyser(OMA) after ALA taked orally( 1h.2h.4h.6h.8h. 10h. 12h.24h.48h). Results.. The results showed that fluorescence power was the strongest in the group of ALA40mg/kg after 1h orally. Conclusion:Orally ALA40mg/kg was suitable dose and 1h after administration ALA was suitable diagnostic time.
