检测人IL-15基因表达的RT-PCR方法的建立

DEVELOPMENT OF A RT PCR METHOD FOR DETECTING HUMAN IL 15 mRNA EXPRESSION

本文依据ＣｅｎＢａｎｋ人ＩＬ１５的ｃＤＮＡ序列，设计３个特异性引物，建立了检测ＩＬ１５基因表达的逆转录ＰＣＲ（ＲＴＰＣＲ）方法。用二次扩增和模板倍比稀释法，证明本方法特异、敏感、简单，初步检测了部分已报道的组织细胞，结果和文献报道的Ｎｏｒｔｈｅｒｎ印迹杂交的结果一致。说明用ＲＴＰＣＲ检测ＩＬ１５ｍＲＮＡ表达是可行的，这为进一步研究ＩＬ１５在人体不同组织中的表达及评价其意义奠定了基础。

Based on the human IL 15 cDNA sequence in GenBank,three primers of PCR were designed,and a RT PCR method of detecting human IL 15mRNA expression was developed.This method showed with very high specificity,sensitivity and simplicity.The results of IL 15 mRNA expression in some normal human tissues were identical with the results of Northern blot assay reported before.This provides foundation to study the expression of IL 15 in different tissues and to evaluate the significances of IL 15.
