摘要
本文对新鲜何首乌根、茎、叶的电子显微镜制样技术进行了探索,结果发现:应用割断扫描电镜ODO法,有利于从不同层次和不同方位分辨该样品的三维超微结构,有利于药用植物的分类鉴定。冷冻蚀刻电镜技术制备生鲜植物样品,由于细胞内含水量大,细胞壁厚,易产生冰晶,该技术尚需改进。
The paper discribed the preparation of freeze etch and SEM techniques for fresh root,stem and leave of Polygonum Multiflorum.The results proved practical value of ODO SEM technique in observing different layers and directions of the herb.It could be helpful to identify the classification of the herb.The SEM technique should be improved since the wall of the cell is thick containing with much water,ice cristal is easily formed.
