摘要
目的:探讨间歇性甲状旁腺激素(PTH)应用促成骨的可能机制。方法:大鼠成骨细胞(ROBs)分为两组培养:①对照组,在每个培养周期(24h)中均不加PTH;②PTH间歇刺激组,仅在前6h的培养液中加PTH。收集第3周期24h末的细胞,应用双向凝胶电泳方法比较两组细胞的总蛋白,选择2个在间歇刺激组表达明显上调的蛋白质点进行基质辅助激光解吸离子化-飞行时间质谱(MALDI-TOF-MS)分析。结果:初步确定这2个蛋白分别为β-肌动蛋白(β-actin)和波形蛋白(vimentin),它们均属于细胞骨架蛋白。结论:在PTH间歇刺激组表达上调的β-肌动蛋白和波形蛋白可能参与了间歇性PTH应用的促成骨过程。
Objective:To explore the anabolic effect of intermittently adminstrated PTH on rat osteoblast cells (ROBs). Methods: The ROBs divided into two groups:①Control (Ctr),the cells were cultured without PTH in an incubation cycle (24 h) ;②Intermittent expoure to PTH(Itm), the cells were cultured with PTH at the first 6 h,but without PTH at the subsequent 18 h. Cellular proteins at 24 h were disolved and seperated in two-dimensional gel electrophoresis (2-DE). Two of the dramatically upregulated proteins were detected by matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MADI-TOF-MS). Results:The two upregulated proteins were β-actin and vimentin. Both of them were cytoskeletal proteins. Conclusion: β-actin and vimentin may play a role in the anabolic effect of intermittent PTH on rat osteoblast cells.
