摘要
通过制备抗华支睾吸虫单克隆抗体，建立和稳定了淋巴细胞杂交瘤技术方法，并对影响融合效果的融合条件进行了观察。聚乙二醇浓度、新生牛血清质量、更换ＨＡＴ培养基的时间等因素对融合效果有影响。筛选了５株分泌高滴度单克隆抗体的杂交瘤细胞株。
The fusion conditions of the hybridoma cells were studied. The results showed that PEG concentration, the quality of new born calf serum and the time for replacing HAT selective media affected the fusion rates. The fusion rates were higher with 50% PEG of MW4000 and replacement of HAT medium on the third day after fusion. The 96 well microtiter plates neednt't to be pre treated. Hybridoma clonies appeared on the third day after fusion. It was usually appropriate to begin to detect antibodies on the seventy day. There were very few new hybridoma clonies to appear after thirteen days. Six fusions were conducted and five hybridoma cell lines secreting high level of monoclonal antibody were screened out.
