脑源性神经营养因子及酪氨酸激酶受体B在大鼠小脑的表达

摘要 目的研究脑源性神经营养因子及酪氨酸激酶受体B在大鼠小脑的时空分布变化。方法采用免疫组织化学方法实验,观察脑源性神经营养因子及酪氨酸激酶受体B在正常大鼠发育期、成年期和老龄期小脑内的定位分布及时间变化。结果发育期小脑皮质的外颗粒层、梨状细胞层和内颗粒层均有BDNF表达,梨状细胞层的阳性Purkinje细胞染色明显。成年期内颗粒层阳性细胞逐渐增多,梨状细胞层的Purkinje细胞BDNF表达及阳性细胞数量达高峰。老年期阳性细胞染色强度呈增龄减低,Purkinje细胞发生了轻度的退行性变,TrkB表达在小脑皮质分布模式类似BDNF的表达。结论小脑皮质BDNF及TrkB的时空表达基本一致。表达高峰为生后20天。老龄期呈增龄性下降,细胞出现退行性改变。 Objective To study systematically the distribute of BDNF and TrkB in the normal rat cerebellum, Methods By immunohistochemistry way tO observe systematically the expression of changes in BDNF and TrkB in the normal rat cerebellum during development phase and adulthood and aged phase. Results BDNF and TrkB were expressed in cerebellum during development phase, The external granule layer became molecular layer along with the migration of granule ceils to the internal granule cell layer, and the immunostaining cells increased gradually during adulthood phase. The Purkinje immunoreactive cells appeared to peak. BDNF and TrkB were expressed significantly in Purkinje cells during adulthood. During aged phase, Purkinje cells slightly degenerated. Conclusions The expression of BDNF and TrkB reached a peak in cerebellum of rats on PND 20, The expression of BDNF and TrkB decreased gradually after adulthood. During aged phase, the expression of BDNF and TrkB increased significantly, and the form of the cell occurred degeneration.
