摘要
用胶原酶消化分离大鼠、猪胰岛,用葡聚糖或聚蔗糖梯度密度离心纯化胰岛并将其包埋于海藻酸钠-聚赖氨酸-海藻酸钠微囊内,用双硫腙染色鉴别胰岛,用荧光染色计数胰岛存活率。将此法制备的微囊胰岛用于治疗Ⅰ型糖尿病有良好的应用前景。
Abstract:Islets of Langerhans were isolated in high yields from rat and porcine pacreata. In the procedure, the pancreata were perfused and digested with collagenase, and the islets were then purified on histopague density gradients. Islets were encapsulated in alginate-polylysine-alginate membranes. Isolated tissue were stained by dithizone to identify islets and were stained by fluorcesin to determine viability. The results of the study indicate a considerable clinical potential of microencapsulated islet xenografts to treat type Ⅰdiabetes.
