摘要 目的探讨uPA、V-ATP酶mRNA在肝细胞癌中的表达及其意义。方法用RT-PCR方法检测45例肝细胞癌及其癌旁组织uPA和V-ATP酶mRNA表达,以7例肝血管瘤作为对照组。结果uPA mRNA在癌组、癌旁组及对照组的阳性率分别为100%、84.4%和28.6%,3组比较差异显著(P<0.05);V-ATP酶mRNA在癌组、癌旁组及对照组的阳性率分别为100%、86.7%、57.1%,3组比较均差异显著(P<0.05);以有/无肝内静脉癌栓形成和肝外转移将癌组分为转移和无转移两组,uPA、V-ATP酶mRNA在两组的表达均有差异显著(P<0.01);而它们的表达与乙型肝炎、病理分级、血清AFP水平以及肿瘤直径无明显关系(P>0.05)。结论肝细胞癌中uPA、V-ATP酶mRNA高表达与肝癌的侵袭转移有关,有可能成为临床治疗的靶点。 Objective To study the mRNA expression of uPA and V-ATPase in human hepatocellular carcinoma and their significance. Methods Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of uPA and V-ATPase mRNA in 45 cases of hepatocellular carcinoma (HCC), their pare-cancer tissues and 7 cases of pare-tumor tissues inhepatic hemangiomas (as control group). Results In cancer, para-cancer and control groups, the positive rate of uPA mRNA were 100%, 84.44%, and 28.57%; the positive rate of V-ATPase mRNA were 100%, 86.67% and 57.14%, respectively. The differences of the expression of uPA and V-ATPase mRNA were significant between three groups respectively ( P 〈 0.05).In addition, the positive rate of uPA, V-ATPase mRNA in HCC were significantly correlated with the portal vein tumor thrombus and the metastasis outside of the iiver (P 〈0.01), but not correlated with the type B hepatitis, pathological stage, the level of serum AFP and the diameter of tumor ( P 〉 0.05). Conclusions The high expression of uPA and V-ATPase mRNA in HCC might be related to the growth, invasion and metastasis of HCC, and they might be used as targets for clinical treatment.
