摘要
目的通过阿齐霉素与二甲酚橙的显色反应来快速测定阿齐霉素中的阿齐霉素含量。方法利用分光光度法通过分光光度计测定制剂中有效成分的含量。结果阿齐霉素与二甲酚橙在乙醇-水介质中发生显色反应,生成了络合物,生成物的颜色发生变化,产物最大吸收波长为582 nm,表观摩尔吸光系数为1.04×10^4L.mol^-1.cm^-1,稳定常数为5.06×10^5,生成络合物的组成比为2∶1,RSD为0.96%(n=8),药物浓度在10-70 mg.L^-1范围内与吸光度值呈线性关系,回收率符合要求。结论此方法简便易行,便于操作,条件易于控制,在实际工作中具有可应用性。
OBJECTIVE To determine the content of azithromycin through the reaction between azithromycin and xylenol orange. METHODS Utilizing the color reaction between azithromycin and xylenol orange in the ethanol-water solution, a spectrophotometic method was used to the determination of Azithromycin content. RESULTS The color reaction between azithromycin and xylenol orange in ethanol-water could form the complex of azithromycin and xylenol orange. The apparent molar absorptivity of complex at 582 nm is 1.04 ×10^4L·mol^-1· cm^-1 , stability constant is 5.06 × 10^5 , the composition of the complex between azithromycin and xylenol orange was 2: 1. The average standard deviation is 0.96% (n =8). Beer's law is obeyed in the range of 10×70 mg· L^-1 of azithromycin in tablets. CONCLUSION The method is simple with easily-controlled reaction conditions and high sensitivity.
