摘要
目的建立高效液相色谱法测定人血浆中甘氨双唑钠浓度。方法以替硝唑为内标,Ultrasphere-ODS C18(4.6 mm×250mm,5μm)色谱柱,以0.02 mol.L^-1草酸铵溶液-甲醇=65∶35,流速为1 mL.min^-1为流动相,检测波长320 nm。结果甘氨双唑钠在5-500μg.mL^-1范围内呈良好的线性关系(r=0.999 7),甘氨双唑钠低、中、高3个浓度的方法回收率大于98%,日内及日间精密度RSD均小于10%。结论本法准确、简便、快速适合于人血浆中甘氨双唑钠浓度的测定。
OBJECTIVE To establish a HPLC method to determine the concentration of glycodidazoliumnatrium in human plasma. METHODS The Ultrasphere-ODS C18 (4.6 mm × 250 mm, 5μm) column was used. A mixture of 0.02 mol · L^-1 ammonium oxalate and methanol (65: 35) at a flow rate of 1 mL · min^-1 was used as mobile phase, and tinidazole was used as internal standard. The detecting wavelength is 320 nm. RESULTS The linear range was from 5 - 500 μg ·mL^-1 ( r = 0.999 7) for glycodidazoliumnatrium. The method recoveries within high and low glycodidazoliumnatrium concentration in blood are more than 98%, the RSD of intraday and inter-day precision are less than 10%. CONCLUSION This method is accurate, simple, rapid, and suitable for the determination of glycodidazoliumnatrium concentration.
