摘要
目的研究肝癌发生过程中原位细胞凋亡活动以及紫杉醇对肝癌细胞系BEL-7404诱导细胞凋亡的作用。方法用脱氧核糖核酸末端转移酶介导的缺口末端标记法检测了肝细胞癌的细胞凋亡,用紫杉醇处理培养细胞BEL-7404、显微镜下进行观察并且进行琼脂糖电泳。结果癌旁组织的原位凋亡细胞增多,紫杉醇处理肝癌细胞可以诱导细胞产生典型的细胞凋亡形态学变化。DNA发生断裂,琼脂糖电泳产生梯形条带。结论在体内肿瘤细胞凋亡降低,细胞分裂可能加快,肿瘤发生。在癌旁组织中细胞凋亡增多,在体外紫杉醇可以诱导肝癌细胞系BEL-7404凋亡。
In order to investigate the action of in situ apoptosis in occurrence of liver carcinomas and the biological effect of taxol on BEL - 7404 cell line inducing apoptosis, Methods- The apoptosis in the hepatoceluller carcinomas was detected with terminal deoxynucl neotidyl transferase mediated dUTP nick end labelling (TUNEL) ; the BEL -7404 cell line cultured in DMED medium were treated with taxol and observed under microscope, and their DNA was assayed by gel electrophoresis. Results: In situ apoptotic cells in the cancer adjacent tissues inceased; treatment of liver cancers with taxol could induce the cells to manifest a typical apoptotic morphology. Their DNA was fractured and a characteristic ladder pattern could be found using electrophoresis. Conclusion: In vivo the apoptosis of carcinomas was lower; maybe the cells divided quickly and then the cancers occurred. In the cancer adjacent tissues. the anontosis nricked un. and in vitro taxol could induce the apoptosis of BEL -7404 cells.
