摘要
目的:探讨含Nell-1基因逆转录病毒液与聚羟基丁酸酯(PLGA)生物材料复合后修复犬下颌骨缺损的效果.方法:制备含Nell-1真核细胞表达载体的逆转录病毒液PT-PLNCX2-Nell-1.制备比格犬下颌骨15mm×10mm×5mm的箱状缺损模型,修复实验分为2组:PT-PLNCX2-Nell-1+PLGA修复组(实验组),PLGA修复组(对照组),每组8个样本,分别于术后8,16wk进行大体观察、X线检测和组织学检测.结果:PT-PLNCX2-Nell-1重组病毒液与生物材料复合修复组可见在骨缺损处有骨性连接形成,骨缺损修复,组织学观察见有大量新生骨组织形成;PLGA修复组未见骨性连接形成,仅在部分骨缺损两断段形成少量骨组织.结论:利用Nell-1基因体内局部、直接转移的方法能够用于犬下颌骨缺损的修复.
AIM: To detect the repairing ability of PT-PLNCX2- Nell-1 viral solution composited with PLGA biomaterials for segmental mandible defect of the beagle dogs. METHODS: Nell-1 eukaryotic expression vector was constructed in vetroviral solution PT-PLNCX2 -Nell-1. Segmental 15 mm × 10 mm × 5 mm defects were created surgically in the mandible of beagles. The PLGA was used alone (as control group, n = 8 ) or in conjunction with PT-PLNCX2-Nell-1 retroviral solution (as experimental group, n = 8 ). Their repairing abilities were assessed by gross observation, X-ray examination and histological detection of the repaired bone. RESULTS: All defects in the experimental group healed radio-graphically at 16 weeks postoperatively,while no bone healing was found in the control group. Histological analysis of the specimens revealed that the defects in the experimental group were filled with coarse trabecular bone, whereas in the control group there was only fibrous tissue filled in the defects. CONCLUSION: Direct, local retroviral delivery of Nell-1 gene may lead to the healing of segmental mandible defect in beagles.
