摘要
目的探讨监测卡马西平血药浓度的方法。方法采用反相高效液相色谱(RP-HPLC)法,反相C18色谱柱(250mm×4.6mm,5μm),流动相为乙腈-甲醇-水(40∶10∶50),柱温35℃,流速0.9mL/m in,检测波长为210nm。结果卡马西平质量浓度在2.5~20μg/mL范围内,与内标的峰高比具有良好的线性关系(r=0.999794),最低检测限为0.109μg/mL,高、中、低3种浓度的平均回收率为95.16%,97.28%和96.37%;精密度试验的日内RSD为2.41%~3.57%(n=5),日间RSD为3.26%~4.02%(n=5)。结论该方法简单、快速、准确,可用于卡马西平的血药浓度监测。
Objective To establish the method of detecting carbamazepine concentration in blood suiting to our experimental condition. Methods HPLC method was used. C18 column was adopted and the temperature was set at 70℃. The mobile phase was a mixture of methanol and water (75:25)and the flow rate was 0.9 mL/min. The UV detector was operated at 220 nm. Results The calibration curve was linear in the concentrations range from 2. 5 to 20μg/mL in blood(r=0.999 794).Within the range, the recovery rate of low, moderate and high concentrations was 95.16%, 97.28%, 96. 37%, respectively. The low limit of detection was 0. 109 μg/mL. The intraday RSD was 2.41% - 3.57% ( n = 5 ). The interday RSD was 3.26% - 4.02% ( n = 5 ). Conclusion This method is easy, convenient and precise to detect the drug concentration in blood of carbamazepine.
