快速分离柱HPLC测定猪肉中氟喹诺酮类兽药的残留 被引量:12

摘要 研究了用快速分离柱高效液相色谱法测定猪肉中7种氟喹诺酮类兽药残留的方法(恩诺沙星、环丙沙星、左氧氟沙星、依诺沙星、诺氟沙星、氟罗沙星及洛美沙星).样品采用1%醋酸/乙腈提取,经c;。固相萃取小柱预分离;以ZORBAX Stable Bound(4.6mm×50mm,1.8μm)快速分离柱为固定相,20%的乙腈(含0.1%的磷酸)为流动相,检测波长为281nm,主要氟喹诺酮类兽药在3.5min内可达到基线分离.待测组分与峰面积在0.2~50μg,/mL内成良好的线性关系,方法检出限达到2.5~4.6ng/mL,加标回收率在95%~104%之间,RSD〈3.2%(n=5)。新方法准确、灵敏、快速. A method for the simultaneous determination of seven fluoroquinolones (Enrofloxacin, Ciprofloxacin, Levofloxacin, Enoxacin, Norfloxacin, Fleroxacin and Lomfloxacin) in pork was developed. The pork sample was extracted by acid -treated acetonitrile and purified by a C18 solid -phase extraction cartridge. Thereafter, the fluoroquinolones were separated on a ZORBAX Stable Bound (4.6×50 mm, 1.8 μm) rapid column with acetonitrile (containing 0. 1% of phosphatic acid) as mobile phase at a flow- rate of 2. 0 mL/min and detected at 281 nm. The seven fluoroquinolones can be separated completely with in 3.5 rain. The limits of detection (LOD) were from 2. 5 to 4. 6 ng/mL. The linearity of calibration curves in concentration range 0.2 to 50 μg/mL were well. The average recovery was 95% - 104% and RSD (n =5) 〈3.2%. This study provided a feasible, accurate and sensitive method for the determination of fluoroquinolones.
