HPLC法同时测定香加皮中杠柳毒苷和4-甲氧基水杨醛的方法改进

Improving of simultaneous determination of periplocin and 4-methoxy salicylic aldehyde in cortex periplocae with HPLC method

[目的]建立同时测定香加皮中杠柳毒苷和4-甲氧基水杨醛含量的方法。[方法]采用高效液相色谱(HPLC)法,ODS色谱柱,流动相为乙腈∶甲醇∶水=27∶10∶63,检测波长220nm。[结果]杠柳毒苷线性范围0.08～1.0μg,r=0.9993;4-甲氧基水杨醛线性范围0.22～3.0μg,r=0.9999;杠柳毒苷和4-甲氧基水杨醛精密度的峰面积(RSD)值分别为0.79%、0.67%,重现性的RSD值分别为1.2%、2.2%,加样回收率分别为97.1%、96.2%。[结论]本法可同时测定香加皮中杠柳毒苷和4-甲氧基水杨醛的含量,分析时间短,且改进方法的准确度和重现性要优于文献报道方法。

[Objective] To establish the method of simultaneous determination of the content of periplocin and 4-methoxysalicylic aldehyde in Cortex periplocae. [Methods] The determination was carried out by HPLC method on ODS chromatographic column, using acetonitrile：methanol：water （27：10：63） as the mobile phase at 220 nm. [Results] The calibration curve was linear within the range of 0.08～1.0 μg, r=0999 4 for periplocin and 0.22～3.0 μg, r=0999 9 for 4-methoxy salicylic aldehyde. The precision of the two constituents was 0.79% and 0.67%, the reproducibility was 1.2% and 2.2% and the recovery rate of samples was 97.1% and 96.2% respectively. [Conclusions] The method can be used to determine pefiplocin and 4-methoxy salicylic aldehyde simultaneously. It is more convenient and accurate than the method reported in the literatures.
