摘要 目的:建立一种改进的利用天然凝胶电泳(N-PAGE)分析多肽间相互作用的方法。方法:主要对上样缓冲液和染色液做了改进。其中,上样缓冲液中除去了SDS和还原剂(DTT或β-巯基乙醇);染色液中加入了0.1%的CuSO4.5H2O,并含有25%的异丙醇。结果:在经过约3h的电泳后,上样量约10μg的多肽(相对分子质量为4000)可与其复合物清晰分离。结论:改进的N-PAGE法可用于研究多肽的相互作用。 Objective: To establish a modified N-PAGE method to determine peptide-peptide interaction. Methods: The loading buffer and staining buffer were improved, in which SDS and reducing agent(DTT or β-mercaptoethanol) were removed and 0.1% of cupric sulfate and 25% isopropanol were used. Results: The peptides with the relative molecular weight of 4 000 Dalton could be separated from their peptide complexes at the loading level of 10 μg after 3 hours run- ning. Conclusion: The modified N-PAGE is a convenient method to determine peptide-peptide interaction.
