摘要 目的研究重组腺病毒介导人乳头状瘤病毒(HPV)16型E7病毒癌基因感染对人宫颈癌HeLa细胞周期蛋白E(Cyclin E)表达的影响,探讨HPV16 E7癌基因以及Cyclin E在宫颈癌发生发展中的作用。方法用含有HPV16 E7病毒癌基因的重组腺病毒载体感染体外培养的人宫颈癌HeLa细胞。免疫荧光染色后激光扫描共聚焦显微镜(LSCM)检测感染前后Cyclin E表达的差异。结果HPV16 E7感染后Cyclin E表达较感染前明显增加。结论HPV16 E7基因的表达促进Cyclin E的表达,对宫颈肿瘤细胞有增殖促进的作用。 Objective To investigate the expression of Cyclin E in HeLa cells after the transfection of HPV16 E7 oncogene by recombinant adenovirus vector, and assess the effect of HPV16 E7 oncogene and Cyclin E in the development of human cervical carcinoma. Methods Expression of Cyclin E was detected by laser scanning confocal microscope (LSCM) before and after the transfection. Results Expression of Cyclin E protein in the HeLa cells increased significantly after the transfection of HPV16 E7 genes. Conclusion Expression of HPV16 E7 oncogene influences the expression of Cyclin E and enhances the proli feration of the cells of cervical carcinoma.
