摘要
目的:探讨热休克蛋白70(HSP70)反义寡核苷酸阻断胶质瘤细胞的HSP70的表达及对胶质瘤细胞凋亡的影响。方法:用MTT法、流式细胞术、琼脂糖凝胶DNA电泳方法观察HSP70反义寡核苷酸对体外培养的胶质瘤C6细胞株HSP70表达的抑制及其对生长和凋亡的影响。结果:用HSP70反义寡核苷酸处理的胶质瘤细胞表现出明显的生长抑制,琼脂糖凝胶电泳显示凋亡细胞DNA降解的特征性DNA阶梯图;MTT法测定的生长抑制率和流式细胞术分析的凋亡率与HSP70反义寡核苷酸呈剂量、时间依赖性关系。结论:HSP70反义寡核苷酸通过抑制HSP70表达而诱导胶质瘤C6细胞株凋亡并抑制其生长,可作为基因治疗胶质瘤的新靶向。
Objective:To investigate the influence of expression of HSP70 and apoptosis of tumor cell with HSP70 antiense oligomer in neuropongioma C6 cells. Methods: The effects of HSP70 antisense oligomer on HSP70 expression levels, the proliferation and apoptosis of neuropongioma C6 cells in vitro were studied by MTT test, flow cytometry and agarose gel DNA electrophoresis. Results: The inhibiting proliferation rate by MTT test and the induced apoptosis rate by flow cytometry analyses revealed the apoptosis and inhibition of C6 cells accurred mainly at G1 and S phase which were dose - and - time dependent. Condution: HSP70 antisense oligomer can induce apoptosis and inhibit the growth of C6 cells by abrogating its HSP70 expression,HSP70 may be used as a target for the gene therapy of neuropongioma carcinoma.
