枸杞、黄芪对大鼠睾丸支持细胞抑制素合成的影响

The Effect of Fructus Lycii and Radix Astragali on Synthesis of Inhibin in Sertoli Cells of Rat Testis

目的：探讨中药枸杞、黄芪对大鼠睾丸支持细胞功能的作用及机制。方法：对18—22d的大鼠睾丸支持细胞进行分离培养,用MTT法检测枸杞、黄芪对支持细胞增殖的影响，用免疫组化方法检测枸杞、黄芪对正常状态和过氧化物（H2O2）损伤状态下支持细胞抑制素βB亚基（INHβB）合成的影响。结果：枸杞、黄芪及杞芪合剂对支持细胞的增殖有促进作用；在正常培养状态下，枸杞、黄芪及杞芪合剂对支持细胞INHβB合成具有促进作用；在过氧化物（H2O2）损伤状态下，枸杞、黄芪及杞芪合剂对支持细胞INHβB合成具有保护作用。结论：枸杞、黄芪及杞芪合剂对体外培养的支持细胞抑制素βB亚基合成具有促进和保护作用。

Objective：To establish in vitro culture of rat Sertoli cells and to investigate the effects of Chinese herbal medicines to their functions and the mechanism. Methods： Sertoli ceils aged 18 - 22 days were separated and cultured from rat testis ; the effect of Fructus Lycii, Radix Astragali and the combined administration of the two medicines on the proliferation of Sertoli ceils in vitro were detected by means of MTT assay; the effect of Fructus Lycii, Radix Astragali and the combined administration of the two medicines on the level of INHβB in Sertoli ceils in vitro were investigated in both normal environment and peroxide-damaging environment by means of immunohistochemistry. Results： The proliferation of Sertoli cells in vitro was promoted when treated with Fructus Lycii, Radix Astragali and the combined administration of the two medicines： Sertoli ceils INHβB was up regulated by Fructus Lycii, Radix Astragali and the combined administration of the two medicines in vitro; Fructus Lycii and Radix Astragali were found to have protective effect on synthesis of INHβB in Sertoli ceils with injury induced by peroxide （H2O2 ）. Conclusion： The function of promotion and protection were found when Sertoli ceils cultured in vitro treated with Fructus Lycii, Radix Astragali and the combined administration of the two medicines.
