摘要
目的探讨健骨二仙提取液对去卵巢大鼠皮质骨形态计量学的影响,进一步研究该药防治骨质疏松症的作用机理。方法将42只6.5月龄雌性近交系大鼠随机分为4组:假手术组、单纯去卵巢组(即模型组)、卵巢切除+健骨二仙提取液(即治疗组)、卵巢切除+倍美力组(即倍美力组),采用去卵巢方法进行骨质疏松造模,6周后药物干预治疗10周,采用皮下注射盐酸四环素和钙黄绿素进行骨表面的双荧光标记,全部处死大鼠取右胫骨中段不脱钙骨切片进行骨形态计量学参数测定。结果与去卵巢组相比较,治疗组骨髓腔总面积减小(P<0.05),外膜面的骨形成各项参数均无明显变化,治疗组和倍美力组骨内膜面的骨吸收参数均降低(P<0.01),治疗组内膜面骨形成参数降低(P<0.05或<0.01),使皮质骨转换水平比倍美力组更接近假手术组水平。结论健骨二仙提取液可抑制去卵巢后高转换型骨质疏松的形成,总体抑制骨吸收大于骨形成,减少了骨量丢失和骨结构退化,是该药防治骨质疏松症的机理之一。
Objective To study the effect of Jiangu Erxian extraction on the histomorphometry of middle tibia cortex in OVX rats and the mechanism of the medicine on preventing osteoporosis. Method The 42 female Wistar rats (WA/Jic, inbred line, SPF grade) were randomly divided into 4 groups: Sham operation group (n=11) fed with normal saline, OVX group (n=10) with normal saline, OVX+herb group(n=11) with the extraction and sham operation+Prematin group(n=10) with premarin. The osteoporosis models were induced by ovariectomied except for group sham operation and treated 10 weeks after 6 weeks of operation, the double fluorescence labeling on the bone surface was prepared with tetracycline-HCl and Calcein hypodemic injection. The right middle tibias were processed by undecalcified section. Then, all the rats were killed and their right undecalcifying tibia sections were detected for quantitative bone histomorphometry. Result Compared with OVX group, the total area of the marrow cavity in therapeutic group was decreased (P〈0.05), all the osteogenic parametes on exterior periosteal surface were not changes, the bone absorbing parametes on interior periosteal surface in therapeutic and premarin groups lowered(P〈0.01) and the osteogenic parametes on interior periosteal surface in therapeutic group lowered (P〈0.05 or P〈0.01). Compared with premarin group, the transforming level of tibia cortex was closed to that in sham group. Conclusion The extraction can inhibit the high transforming type of osteoporosis and reduce the losing speed of bone mass and prevent osteoporosis, and the total bone absorption is higher compared with the bone formation.
