摘要
目的研究美罗培南对铜绿假单胞菌PAO1野生型及其lasR/rhlR基因缺陷型菌株生物被膜的作用。方法采用生理盐水-吸痰管系统进行铜绿假单胞菌PAO1野生型、PAO1lasR、PAO1 rhlR基因缺陷型体外生物被膜培养7d,经26μg/ml美罗培南作用24h,扫描电子显微镜观察生物被膜的变化。结果7d后PAO1野生型和PAO1lasR、PAO1 rhlR基因缺陷型都能形成生物被膜结构,美罗培南作用后,PAO1野生型的生物被膜结构明显稀薄,而PAO1lasR、PAO1 rhlR基因缺陷型的生物被膜结构基本被破坏。结论lasR,rhlR基因抑制美罗培南对P.a生物被膜内的敏感性。
Objective To understand the effect of meropenem on the biofilms of P. aeruginosa lasR/rhlR mutantions in vitro. Methods The biofilms of PAO1 and lasR, rhlR mutants were incubated in a polyvinyl chloride tube in 0.9% saline for seven days,and then they were immersed in meropenem solution of 26μg/ml for 24 hours, followed by examinations of scanning electronic microscopy (SEM). Results After 7 day's incubation, PAOI and lasR, rhlR mutants biofilms showed a well-developed structure;after 24 hours effect by meropenem, PAO1 biofilm become thin and poor,whereas lasR, rhlR mutants biofilms were almost destructed and there were some small pellicle-shaped ones. Conclusion lasR/rhlR genes play probably an important role in the destructive effect of meropenem on the P. aeruginosa biofilms in vitro.
