离体玉米精细胞保存及其呼吸代谢的初步研究

摘要 对离体玉米精细胞的保存与代谢的研究结果如下:利用二步渗透压冲击法分离玉米精细胞,产率为3×105个/mL,FDA(Fluoresceindiacetate)荧光反应活性为90%以上;在一定保存介质中,精细胞在常温下72h可存活40%;离体精细胞具有正常呼吸代谢,其有氧呼吸80%以上为线粒体呼吸,随着保存时间延长,呼吸强度下降。 Studies have been conducted on the following aspects: Sperm cells were isolated from maize(Zea mays L) pollen grains by two-methods of osmotic shock, and the isolated sperm cells were concentration 3×10 5 cells/mL, the viability had been testd by using fluorochromatic reaction(FCR) was over 90%; Tested by FDA the isolated sperm cells stored at 20 ℃, 40% cells survived 72 hours; The sperm cells exsist normal respiration, and over 80% of them belongs to mitochondria respiration by using inhibitor. During their presevation, the respiratory rate gradual decreased.
