摘要
目的研究对小鼠不同频率的重复经颅磁刺激(Repetitive transcranial magnetic stimulation,rTMS)的抗抑郁作用,并探讨重复经颅磁刺激的安全性。方法昆明种小鼠分为高频组、低频组、模型组及对照组。高频组和和低频组分别接受连续5天的高频重复经颅磁刺激(10Hz)和低频重复经颅磁刺激(2Hz)处理,模型组与对照组不接受rTMS处理。以开场实验研究rTMS的安全性,以强迫游泳实验研究不同频率rTMS对小鼠抗抑郁样作用,同时通过海马肾上腺素受体对其机制进行探讨。结果研究发现在开场实验中,高频组、低频组和对照组小鼠的行为指标,包括:穿越格数,直立数,理毛数和排泄数差异均无显著性。在强迫游泳实验中,与模型组相比,低频组的潜伏期延长,悬浮时间缩短,同时不同频率的重复经颅磁刺激对小鼠海马肾上腺素受体的表达产生不同的影响。结论不同频率的重复经颅磁刺激的抗抑郁样作用不同,低频重复经颅磁刺激具有一定的抗抑郁样作用。而高频和低频的重复经颅磁刺激均具有安全性。
Objective To investigate the antidepressant-like effect and security of different frequency of repetitive transcranial magnetic stimulation (rTMS). Methods Thirty mice were divided into high frequency group, low frequency group ,control group and model group. The high frequency group and the low frequency group received rTMS for 5d and the control group and model group were not treated with rTMS. The open field test and force swimming test were performed to observe the security and antidepressant effect of rTMS, respectively, and the expression of mRNA of adrenalin receptors in hippocampi was detected with RT-PCR. Results Open field test displayed no significant difference in the high/low frequency group and control group. Force swimming test showed the latency of low frequency group increased while the immobility reduced compared with model group. The expression of mRNA of adrenalin receptors in hippocampi in low frequency group was similar to that in control group, but lower than that in high frequency group. Conclusion rTMS within certain frequency and intensity is safe. Low frequency of rTMS has antidepressant-like effect.
