骨愈1号药物血清对成骨细胞增殖分化功能的影响 被引量:2

摘要 目的:观察中药骨愈1号药物血清对成骨细胞增殖、分化以及矿化的影响。方法:从新生SD大鼠颅骨获取成骨细胞进行培养,取第3代成骨细胞进行试验,将其分为4组,在各组培养液中分别加入不灌胃骨愈1号的新西兰兔血清(对照组)和灌胃骨愈1号浓度分别12.5、25、50g/kg的新西兰兔血清(高、中、低剂量组)。用MTT法测定细胞增殖功能;用对硝基苯基磷酸二钠法测定碱性磷酸酶(ALP)活性;用茜素红染色法,在40倍光镜下作矿化结节计数。结果:与对照组比较,骨愈1号药物血清高、中、低剂量组在MTT显色的OD值、ALP活性和矿化结节数均明显升高(P<0.01或者P<0.05),且高剂量组优于低剂量组(P<0.01或者P<0.05),在提高ALP活性方面,高剂量组优于中剂量组(P<0.05)。结论:骨愈1号具有促进成骨细胞增殖、分化及矿化作用,且其作用呈剂量相关性。 Objective : To observe the effect of drug sermn with No1 Guyu on skeletogenous cell proliferation and differentiation. Methods : The skeletogenous cells which were taken from cranial bones of SD rats cultivated, and were derided into 4 groups, the culture solution of which were added in order rabbit serum without No1 Guyu, and with No1 Guyu of density 12.5g/Kg, 25g/Kg, 50g/Kg. The cell proliferation ability were determined by MTT; the activity of ALP were determined by Nitrophenyl disodium phenylphosphate; the number of mineralization tuberculum were determined by light microscope after alizarin Bordeaux staining. Results : Compared with model group, the value of OD, the number of mineralization tuberculum and the activity of ALP of drug serum with No1 Guyu in high, middle, low - dose groups increased significantly( P 〈0.05 or P 〈0.01 ), and compared with low - dose group group, those of high - dose group decreased significantly( P 〈 0.05 or P 〈 0.01 ) ; the activity of ALP of high - dose group is more improved than those of middle - dose group ( P 〈 0.05). Conclusion : No1 Guyu could improve proliferation and differentiation of skeletogenous cells,which relative to dosage of drug.
