肉苁蓉多糖对骨髓抑制性贫血小鼠造血调控的实验研究 被引量:18

摘要 目的:观察肉苁蓉多糖对骨髓抑制贫血小鼠造血功能的影响,探讨其造血调控作用机制。方法:采用CO60照射和注射环磷酰胺复合制备贫血小鼠模型。应用流式细胞术(FCM)检测肉苁蓉多糖对骨髓抑制贫血小鼠骨髓细胞周期的影响,采用造血祖细胞培养术观察三系造血祖细胞集落数。结果:低剂量和高剂量肉苁蓉多糖能明显促进骨髓G0/G1期细胞向S期细胞以及S期细胞向G2/M期细胞的转化,G2/M期细胞比例明显升高,增殖指数(PI)也明显升高。给药组CFU-E,BFU-E,CFU-MeG与贫血对照组集落数有显著差异。结论:肉苁蓉多糖可能通过促使骨髓抑制贫血小鼠骨髓细胞细胞周期的转化,促进骨髓造血功能的恢复,并促进红系巨核系造血。 Objective: to investigate the effect of Desertliving Cistanche Polysaccharides on haematogenesis function in Bone Marrow Depressed Anemia Mice and explore its mechanism for hematopoietic regulation. Methods: built the mold of anemia mice by irradiated Co^60 and injected cyclophosphamide, The effect of Desertliving Cistanche polysaccharides on flowcytometry (FCM) respectively and cultured hemopoietie progenitor cell to observe three strain of hemopoietic progenitor cell colony number. Results : It was found that low - dose and high - dose Desertliving Cistanche polysaccharides promoted G0/G1 - S phase and S - G2/M phase transition of BMCs, obviously increased the proliferation index and the ratio of G2/M phase cells o Additionally,compared with the control group, the level of CFU -E, BFU- E, CFU -MeG colony number of Desertliving Cistanche polysaccharides group was significant. Conclusion : The data suggest that Desertliving Cistanche Polysaccharides may promote the bone marrow cellcycle transition and the recovery of Hematopoietic function in bone marrow depressed anemia mice, accelerate haematogenesis in rubrum strain and macronucleus strain.
