摘要
目的探讨去细胞牛颈静脉血管经氨等离子表面改性后接枝甘氨酸-精氨酸-甘氨酸-天冬氨酸-丝氨酸-脯氨酸-赖氨酸(GRGDSPK)肽的可行性及效果。方法A组:去细胞牛颈静脉经过氨等离子预处理后与GRGDSPK肽在液相溶液中作用;B组:去细胞牛颈静脉直接与GRGDSPK肽在液相溶液中作用。连续震荡漂洗条件下,激光共聚焦显微镜观察0 h、4 h GRGDSPK肽在2组材料表面的分布特征。结果在溶液作用下,随时间延长,氨等离子处理接枝荧光GRGDSPK-FITC肽的牛颈静脉血管表面肽荧光未见丧失;而物理吸附GRGDSPK-FITC肽荧光显著丧失。结论氨等离子表面改性技术用于生物衍生材料去细胞牛颈静脉可取得良好的效果。
Objective To investigate the possibility that immobilizing bioactive organic molecules Gly-Arg-Gly-Asp-Ser- Pro-Lys-FITC (GRGDSPK-FITC) onto acellular bovine jugular vein (BJV) matrix treated with ammonia plasma. Methods Group A:after treated with ammonia plasma, the vein matrixes were submerged into GRGDSPK-FITC aqueous solution. Group B: the vein matrix was straightly submerged into GRGDSPK-FTTC aqueous solution. The samples of two groups were rinsed continuously. At the time of ohour and 4 hour, the surface fluorescence distribution feature was observed by confocal microscopy. Results Ammonia plasma treatment could effectively holded on the fluorescence of GRGI)SPK-FTTC on the surface of BJV under aqueous solution;Conversely, the fluorescence of GRGDSPK-FTTC attenuated sharply on the surface of BJV by simple adsorption. Conclusion Ammonia plasma is an effective way to immobilize bioactive molecules onto the surface of acellular bovine jugular vein matrix.
