Survivin在喉鳞状细胞癌组织中的表达及与caspase-3、P53相关性的初步研究 被引量:8

摘要 目的研究凋亡抑制基因Survivin蛋白在喉癌中的表达及与caspase-3、P53表达的相关性。方法采用免疫组化SP方法对46例喉鳞状细胞癌组织和16例正常喉黏膜组织中的Survivin蛋白与caspase-3蛋白、P53蛋白进行检测。结果在喉癌组织中Survivin的阳性表达率为71.7%,其阳性表达率与临床分期、肿瘤的分化程度以及淋巴结转移无关;P53阳性表达率为65.1%,caspase-3阳性表达率为26.1%。在正常组织中Survivin蛋白与P53蛋白均不表达,但caspase-3蛋白全部表达;caspase-3与Survivin表达显著负性相关,P53与Survivin表达显著正性相关。结论Survivin在喉癌中高表达提示该基因在喉癌的发生发展中有着重要的作用,caspase-3的失表达、P53的突变与Survivin的阳性表达可能在喉癌的形成过程中起着协同作用。 [Objective] To study the expression of survivin and its relationship with expression of easpase-3, P53 in laryngeal eareinomas. [Method] Immunohistoehernieal assay was used to deteet the expression of Survivin,easpase-3 and P53 proteins in 46 eases of laryngeal squamous eel] eareinomas and 16 normal laryngeal epithelium tissues. [Results] Survivin protein was deteeted in 33 of 46(71.7%) laryngeal eancerous tissues, but no expression of Survivin was deteetable in corresponding normal epithelium tissues. The expression of Survivin was not related to the elinie stage of tumor, differentiation and lymph node metastasis. The easpase-3 protein expression was deteeted in all normal laryngeal epithelium, but only 26.1% positive expression in laryngeal eareinoma. P53 protein was deteeted in 30 of 46(65.1%) laryngeal eaneerous tissues, but no expression of P53 was deteetable in eorresponding normal epithelium tissues. There was negative significant correlation between the expression of Survivin and easpase-3, but positive significant correlation between the expression of Survivin and P53. [Conclusion] The expression of Survivin in laryngeal cancerous tissues is up-regulated, indicating this gene may play an important role in the process of car- cinogenesis of laryngeal carcinoma. The changes in the level of Survivin, caspase-3 and P53 may play important roles in the pathogenesis of laryngeal carcinoma.
