周围神经损伤后外源性人胰岛素样生长因子-1在骨骼肌和脊髓运动神经元细胞的表达

摘要 目的观察周围神经损伤局部转染外源性人胰岛素样生长因子-1(hIGF-1)基因在骨骼肌和脊髓运动神经元细胞的表达。方法90只雄性Wistar大鼠,建立坐骨神经挤压伤模型后随机分为3组。hIGF-1治疗组:挤压伤处神经外膜下即刻注射pcDNAhIGF-1和LipfectAmine转染液10μL;模型组注射pcDNA3.1、LipfectAmine和蒸馏水混合液10μL;空白对照组:不注射任何物质。术后免疫组化法观察不同时相hIGF-1在骨骼肌和脊髓的表达。结果hIGF-1在周围神经损伤局部转染可以通过轴浆运输在骨骼肌和运动神经元细胞表达,表达高峰分别在转染后1周和2周左右。结论周围神经损伤局部hIGF-1转基因能在骨骼肌和脊髓运动神经元细胞有效地表达。 [Objective] To observe the expression in the skeletal muscle cell and dynamoneure cell by partially transfeeting the exogenetie human insulin-like growth factor-1 (hIGF-1) gene when the peripheral nerve injuries. [Methods] 90 Wister male rats with crush pattern of sciatic nerve were randomly divided into three groups, Group MGF-1: 10μL of LipfectAmine-mediated hlGF-1 plasmid was locally injected in sciatic epineurium of the crush spot. Model group: 10μL of LipfeetAmine, pcDNA3.1 and distilled water was given in epineurium of the crush spot. And Blank control group:nothing was given. After operations, we observed the expression of the MGF-1 in skeletal muscle and spinal cord at different time with the immunohistoehemieal methods. [Results] The partial transfeetions of hIGF-1 gene in the peripheral nerve injuries can express in the skeletal muscle cell and dynamoneure cell via axoplasmie transporting, and the respective expression peak appears about one week and two weeks after the transfee- tion. [Conclusion] Partially transfeeting the exogenetie hIGF-1 gene can express effectively to the skeletal muscle cell and dynamoneure cell when the peripheral nerve injuries.
