摘要
目的检测胃癌组织中基质金属蛋白酶-2(MMP-2)和E-钙黏附素(E-cadherin)的表达,探讨其与胃癌侵袭转移的关系。方法对我科2003年12月至2004年4月30例胃癌手术患者,采用免疫组化染色检测胃癌组织中MMP-2和E-cadherin的表达情况。结果70%(21/30)的胃癌组织中MMP-2表达阳性。MMP-2表达阳性与浸润深度(P=0.022)、淋巴结转移(P=0.030)和肿瘤分化程度(P=0.043)密切相关,但与性别、年龄、肿瘤位置和直径、Lauren分型及淋巴管侵袭无关(P〉0.05)。46.7%(14/30)的胃癌组织中E-cadherin表达阴性。E-cadherin表达阴性与Lauren分型(P=0.026)、肿瘤分化程度(P=0.011)、淋巴结转移(P=0.038)和淋巴管侵袭(P:0.001)密切相关。结论胃癌组织中MMP-2表达上调和E-cadherin表达下移可能参与肿瘤侵袭转移的发生。
Objective To detect expressions of matrix metalloproteinase-2 (MMP-2 and E-cadherin in gastric cancer tissues and evaluate their relationship with tumor invasion and metastases. Methods Immunochemical technique (EliVisionTM plus) was used to detect expressions of MMP-2 and E-cadherin in tumor tissues from 30 cases with gastric carcinoma admitted into our hospital from December 2003 to April 2004. Results Expression of MMP-2 was positive in 21 specimens (70%) and negative in 9 (30%). Positive expression of MMP-2 closely correlated with infiltration depth ( P =0.022), lymph node metastasis ( P =0.030) and tumor differentiation ( P =0.043) but had none relation with age, gender, tumor location, tumor diameter, Lauren classification and lymphatic invasion ( P 〉 0.05). Of 30 specimens, 14 (46.7%) had negative expression of E-cadherin, which was significantly associated with the Lauren classification ( P = 0. 026 ) , lymph node metastasis ( P -- 0.011 ), differentiation grade ( P =0.004) and lymphatic invasion ( P =0.001 ). ConclusionPositive expression of MMP-2 and negative expression of E-cadherin may contribute to development of tumor invasion and metastases of gastric carcinoma.
