摘要
目的了解E—selectin、Integrin β1、ICAM-1在人胃癌细胞中的表达水平,探讨胃癌细胞、正常胃上皮细胞与人脐静脉内皮细胞的黏附力学特性及其黏附分子的作用机制。方法采用Northem印迹法和ELISA法检测胃癌细胞、正常胃上皮细胞和人脐静脉内皮细胞E—selectin、IntegrinBl及ICAM-1表达水平;采用微管吸吮技术分别测量了胃癌细胞、正常胃上皮细胞与人脐静脉内皮细胞间的黏附特性,利用抗体阻断实验验证胃腺癌细胞与人脐静脉内皮细胞黏附行为中E—selectin、Integrin131及ICAM-1的作用。结果胃癌细胞、正常胃上皮细胞和人脐静脉内皮细胞均有E-selectin、Integrin131及ICAM-1表达,但胃癌细胞3种黏附分子表达水平均高于正常胃上皮细胞和人脐静脉内皮细胞,差异有统计学意义(P〈0.05)。胃癌细胞与人脐静脉内皮细胞的黏附力值为(31±4)×10^-10N,较之正常胃上皮细胞黏附力明显升高[(20±3)×10^-10N],两者差异有统计学意义(P〈0.001)。抗体阻断前后细胞之间的黏附力比较差异有统计学意义(P〈0.05)。结论E—selectin、Integrin β1及ICAM-1可能与胃癌细胞转移有关。
Objective To understand expressions of E-selectin, Integrin β1 and ICAM-1 in gastric cancer and investigate the adhesive mechanics of gastric cancer cells and normal stomach epithelium to umbilical vein endothelial cells as well as the mechanism of adhesive molecules. Methods Nothern Blotting hybridization and ELISA were used to detect expressions of E-selectin, Integrin β1 and ICAM-1 in gastric cancer cells, normal stomach epithelium and umbilical vein endothelial cells. In the meantime, the adhesive properties of gastric cancer cells and normal stomach epithelium with umbilical vein endothelial cells were observed by using technique of micropipet aspiration. Then, the roles of E-selectin, Integrin β1 and ICAM-1 in adhesiveness of gastric cancer cells with umbilical vein endothelial cells were proved by antibody blockage experiment. Results In gastric cancer cells, normal stomach epithelium and umbilical vein endothelial cells were found expressions of E-selectin, Integrin β1 and ICAM-1, which were higher in gastric cancer cells than those in normal stomach epithelium and umbilical vein endothelial cells, with statistically significant difference ( P 〈 0.05 ). The adhesive forces of gastric cancer cells and normal stomach epithelium to umbilical vein endothelial cells were (31 ±4) × 10^-10 N and (20 ± 3) × 10^-10N respectively, with statis tically significant difference between two groups ( P 〈0.001 ). There was statistical difference upon adhesive forces of gastric cancer cells to umbilical vein endothelial cells before and after blockage ( P 〈 0.05 ). Conclusion E-selectin, Integrin β1 and ICAM-1 may be related to tumor metastasis.
