环丙沙星滴眼剂在家兔眼内组织分布及其药物动力学

Distribution and pharmacokinetics of topically applied ciprofloxacin eye drop in the rabbit eye

环丙沙星（ｃｉｐｒｏｆｌｏｘａｃｉｎ，ＣＰＦＸ）滴眼液点入兔眼后，用ＨＰＬＣ法测定眼内各组织中药物浓度。结果在角膜、房水、虹膜－睫状体、晶体、玻璃体内峰浓度值分别为１９．４３μｇ／ｇ，１．５８μｇ／ｍｌ，１６．６８μｇ／ｇ，１．４２μｇ／ｇ和０．９６μｇ／ｍｌ；其半衰期分别为０．７６，０．６９，０．９２，０．６１和１．４０ｈ。结果表明ＣＰＦＸ能在眼内达到较高的抗菌浓度。

The concentration of ciprofloxacin(CPFX) was measured successfuly by HPLC after topically applying 0.3% CPFX eye drops to rabbit eyes.The results showed the CPFX could rapidly penetrate into rabbit ocular tissues. Among all tissues,concentration of CPFX in cornea was the highest,next concentration order was irisciliary body,aqueous humour,lens and vitreous body.The half life(T 1/2 Ke) of the tissue concentration of CPFX in larimal fluid,cornea,aqueous humour,iris ciliary boldy,lens and vitreous body was 0.81h , 0.76h , 0.69h , 0.92h , 0.61h and 1.40h ,respectively.The peak concentration (C max ) of CPFX in the tissue was 19.43 μg/g in cornea, 1.58 μg/ml in aqueous humour, 16.68 μg/g in iris ciliary body, 1.42 μg/g in lens and 0.96 μg/ml in vitreous body.The experiment indicates that higher CPFX antibiotic concentration in various ocular tissues could be observed.
