一氧化氮合成酶mRNA转录强度对妊高征的影响 被引量:2

The effects of iNOS-mRNA transcription level on pregnancy induced hypertension.

摘要 采用窄线杂交的方法测定妊高征患者及正常晚孕妇女子宫及胎盘组织中iNOS-mRNA的转录强度;并用Greiss方法测定子宫及胎盘中NO-2/NO-3、cGMP含量。结果:妊高征患者子宫及胎盘组织中iNOS-mRNA的转录强度明显降低(P<0.05);NO-2/NO-3及cGMP含量亦显著下降(P<0.01);两种组织中NO-2/NO-3含量与cGMP含量呈正相关(P<0.001)。结论:内源性舒血管因子NO合成的减少,可能是妊高征发病过程中的一个重要环节;妊高征时组织中iNOS-mRNA转录水平下调,iNOS生成减少,是NO合成减少的重要原因之一。 Objective:The study was initiated to determine whether there were changes of iNOS-mRNA transcription level and nitric oxide(NO) content in uteri and placentas in patients with PIH.Design:14 patients with PIH and 14 health pregnant women in the third trimester were studied.The levels of iNOS-mRNA transcription in uteri(n=6) and placentas(n=5)were detected with the Dot Blot.The contents of NO 2 -/NO 3 -,the stable metabolic end products of NO,and cGMP in uteri(n=14) and placentas (n=14) were measured with the Greiss reagent.Results:①The levels of iNOS-mRNA transcription in uteri and placentas in the patients with PIH obviously declined compared with health pregnancy women(P<0.05).②The contents of NO 2 -/NO - 3and cGMP in uteri and placentas in the patients with PIH apparently decreased compared with health pregnancy women(P<0.05).③In both uteri and placentas,there was a positive correlation between the content of NO 2 -/NO 3 -and the contents of cGMP(P<0.001),respectively.Conclusion:①The decrease of NO synthesis may be one of the important factors responsible for PIH.②One of the possible reasons for the decrease of NO synthesis in patients with PIH is the reduction of iNOS synthesis induced by the drop of iNOS-mRNA transcription. \ \
