摘要
目的考察活性保护剂对微球中蛋白活性的保护作用。方法以超氧化物歧化酶(SOD)为模型药,采用W/O/W复乳-溶剂挥发法制备SOD-PLGA微球,采用BCA蛋白浓度测定试剂盒测定SOD蛋白浓度,黄嘌呤氧化酶系统测定SOD活性,考察多糖、多元醇、表面活性剂等活性保护剂对SOD活性的保护作用,并探讨保护剂之间是否有联合作用及保护剂含量对SOD活性保护作用的影响。结果活性保护剂对SOD活性均有保护作用。BSA和β环糊精有协同作用。在一定范围内,随着保护剂用量的增加,活性保护作用增强。结论BSA和β环糊精较好的保护了PLGA微球中SOD的活性。
OBJECTIVE To investigate the effect of activity protectors on the protein activity in microspheres. METHODS Superoxide dismutase(SOD) was chosen as the model protein. SOD-PLGA microspheres was prepared by W/O/W multiple emulsion technique. The concentration of SOD was determined by BCA and enzyme activity was monitored by pyrogallol autooxidation. RESULTS The results of enzyme activity showed that all protectors protected the activity of SOD. BSA and β-cyclodextrin had synergistic effect on the protection of SOD activity. And the protecting effect was increased with increasing of the protectors concentration to some extent. CONCLUSION PLGA microspheres loaded with SOD can be protected by some activity protectors. And there are synergistic effect between BSA and β-cyclodextrin,
