^(60)Co照射对MGC-803细胞株癌基因表达的影响

摘要 [目的]观察60Co照射对胃癌MGC-803细胞株癌基因表达的影响.[方法]利用常规方法复苏并培养MGC-803胃癌细胞株,利用60Co照射癌细胞株,在倒置显微镜下观察细胞形态学变化,利用免疫组织化学及原位杂交方法显示癌基因,观察间隙为6 h.[结果]60Co照射前MGC-803胃癌细胞株c-fos,c-ha-ras基因表达稳定;照射6 h后培养瓶内细胞被破坏或变形,且不表达癌基因,12~24 h时部分癌细胞开始表达癌基因,48 h时多数癌细胞表达癌基因.[结论]60Co照射在癌细胞内作用靶点是癌基因的DNA链,被照射后DNA易被破坏使肿瘤细胞的增生被抑制. OBJECTIVE To investigate the effects of ^60Co radioactive ray on cancer gene expression in MGC-803 cells. METHODS The adenocarcinoma MGC-803 cells were recovered and cultured using common method. The cells in the cultured bottle were radiated by ^60Co and the pathological changes were observed using upside-down microscope. The changes of gene expression were examined by immunohistochemistry and in situ hybridization every 6 h. RESULTS The stabilities of c-los and c-ha -ras expression were observed before radiation. The cells were desolate and the gene expression could not examined 6 h after radiation. The gene expression was located in partial cells and out of cells 12-24 h after radiation. The gene expression was seen in most of cells and out of cells. CONCLUSION The target that radioactive ray acts on the tumor is DNA, the DNA was easily damaged after radiation and renewed soon, which makes tumor recurred and transferred partially. The radiontherapy of irradiation means and dosage should be changed and it is necessary to prevent the resurrection of damaged DNA if preventing the recurrence and transference.
