摘要
目的:应用 Hitachi L-8800型氨基酸分析仪对重组人内皮抑素(rh-endostatin)进行氨基酸组成分析,以为其结构确认提供依据。方法:采用酸水解,以外标法测定样品的氨基酸含量及其组成。结果:测定了 rh-endostatin 样品中15种氨基酸的组成,与其理论值相符,误差在15%以内。结论:以氨基酸自动分析仪对样品进行氨基酸组成分析,重复性好,误差较小,能够为样品的结构确认提供一定的依据。
Objective：To analyse amino acid components of recombinant human endostatin （rh -endostatin） so as to provide basis for the characteristic structure by Hitachi L - 8800 amino acid analyzer. Method： Hydrochloric acid hydrolysis, external reference method for determining amino acid content and composition of sample. Result：Determined the 15 kinds of amino acid composition of rh - endostatin samples, the measured value was identical with the theoretical value and experimental error was within 15%. Conclusion： Analyzing sample by amino acid analyzer is reproducible, accurate and can provide foundature for structure identification.
