摘要
将菌毛化的鸡大肠杆菌国内分离株GL7(O78)灭活后作为免疫原,经淋巴细胞杂交瘤技术,用间接酶联免疫吸附试验(ELISA)筛选,获得3E1、3H1、4A9和5G4共4株能稳定分泌1型菌毛抗体的杂交瘤细胞,它们均为IgG。免疫印迹试验结果表明:4株单抗均能同鸡大肠杆菌1型菌毛反应,识别分子质量为17-17.5 ku的菌毛蛋白。同时对鸡大肠杆菌表达1型菌毛的分离株进行检测,结果表明同一血清型或不同血清型间的鸡病原性大肠杆菌1型菌毛之间存在差异,O血清型不能完全体现菌株间的遗传相关性。
Avian Escherichia coli isolated strain GL7 (O78), was inactivated as immunogen to prepare specific monoclonal antibody. 4 strains of monoclonal antibody hybridoma cells were obtained by indirectly enzyme-linked immunosorbent assay (ELISA). They are all IgG, all the 4 strains were specific to the type 1 pill through Western-blotting, and recognized the protein of fimbriae which MW about 17 - 17.5 ku. There antibodies were used as probe to detect the type 1 from Escherichia coli isolated strain from chicken, ware in the same serotype or not, the result showed the there ware exist differences whether there and O serotype can't fully embody the hereditary relativity of different strains.
