摘要
以2个辣椒(Capsicum annuum L.)自交系子叶诱导产生的愈伤组织为材料,辣椒枯萎菌粗毒素为选择剂,筛选抗枯萎病辣椒新种质.结果表明,枯萎菌粗毒素对辣椒子叶愈伤组织诱导、生长及不定芽分化具有明显的抑制作用,且随着粗毒素浓度的增加,抑制作用增强;在枯萎菌粗毒素质量浓度为0.60 g·L-1条件下,筛选、鉴定并获得了抗枯萎病辣椒体细胞变异无性系,且成功再生抗性植株.从而证明,离体筛选抗枯萎病辣椒新种质的方法是可行的.
New pepper (Capsicum annuum L. ) germplasm resistant to fusarium wilt was obtained in vitro. Callus was induced by cotyledon explants under crude toxin, that was extracted from Fusarium oxysporum ,selection stress in two pepper self-inbred lines. The result showed that the crude toxin had clearly negative effect on callus inducement and growth. It also extremely inhibited differentiation of adventitious buds. Some somaclonal variation lines that were selected at the concentration of 0.60 g · L^-1 of crude toxin could resist the crude toxin and regenerate seedlings resistant to fusarium wilt. It was practicable to select new pepper germplasm resistant to fusarium wilt in vitro.
