摘要
采用细管法冷冻保存山羊精液,以甘油作为冷冻保护剂,探讨了加入不同浓度的甘油以及在不同时间加入甘油对精液冷冻效果的影响。结果表明:(1)在稀释液中加入甘油6%和7%,精液的冻后活率与复苏率均显著高于加入3%和5%组(冻后活率:0.49±0.75,0.46±0.80 vs 0.20±0.56,0.31±0.86,P<0.05;复苏率:0.65±0.80,0.61±0.56 vs 0.27±0.65,0.41±0.42,P<0.05),以添加6%的甘油的精液冷冻效果较好。(2)在配液时加6%甘油的处理组在冻后活率与复苏率都显著低于在4°C平衡后加甘油处理组(0.28±0.82 vs 0.34±0.67;0.37±1.0 vs 0.45±0.95,P<0.05)。说明在山羊精液冷冻保存中,加入甘油的时间为4°C平衡后冻结前为较好。
The effects of different concentrations of glycerol, different time of glycerol added into extender were studied, The results were as follow: (1) Post-thawing sperm mobility and recovery rate of sperm in 6%, 7% glycerol were higher than that of 3% and 5% of glycerol significantly (postthawing sperm mobility; 0. 49±0.75.0.46±0. 80 vs 0. 20±0. 56,0.31±0.86,P〈0.05; recovery rate of sperm: 0.65±0.80, 0.61±0.56vs 0.27±0.65, 0.41±0.42, P〈0.05). Among of this, 6% of glycerol in goat extender could have better cryoproteetive ability. (2) When 6% glycerol adding into extender, both of post-thawing sperm mobility and recovery rate of sperm were higher than that of glycerol adding into the extender after semen undergone balance in 4℃ (0. 28±0. 82 vs 0.34±0.67; 0. 37±1. 0 0 vs 0. 45±0. 95, P〈0. 05) . This indicated that it is better time to add 6% of glycerin into the ram extender after semen finishing balance in 4℃ and before freezing.
