摘要
重组菌BL21(pFSFaeG)是能够同时表达致PWD和ED大肠杆菌保护性抗原(F4、F18、Stx2e)基因的基因工程重组菌。由IPTG诱导的重组菌制成抗原,经甲醛灭活后腹腔免疫小鼠,用间接ELISA法定期检测小鼠血清中的抗体产生情况,结果显示重组菌能够诱导小鼠产生针对上述3种保护性抗原的特异性抗体IgG。免疫攻毒保护试验表明,用大肠杆菌标准强毒株C83907(F4)、108/86(F18)和Stx2e毒素攻毒后,重组菌诱导产生的抗体能够对小鼠提供80%以上的保护。
The recombinant strain BL21(pFSFaeG) expressing the subunit protein of F4,F18 and Stx 2e was constructed and its antibody response was detected in mice. The result indicated that serum IgG specific to F4,F18 and Stx 2e antigens were detected in mice immunized with the inactivated vaccine. The vaccinated mice with inactivated recombinant strain can resist the challenge of strain 107/86,C83907 and Stx 2e toxin with the protective efficacy of 81.8%,87.5% and 81.3% respectively.
