脑脉Ⅱ号胶囊抑制脑出血后脑水肿的机理研究 被引量:3

摘要 【目的】探讨脑脉Ⅱ号胶囊(由人工牛黄粉、水牛角、龙胆草、虎杖、水蛭、益母草等组成)对实验性脑出血大鼠凝血酶受体即蛋白酶激活受体-1(protease-activated receptors-1,PAR-1)表达的影响。【方法】将104只大鼠随机分为正常组(N=8)、模型组(N=32)、脑血康组(N=32)和脑脉Ⅱ号组(N=32);除正常组外,其他3组分别于造模后6 h、24 h、3 d、7 d,采用免疫组化法测定不同时间点大鼠脑出血后血肿周围水肿组织中PAR-1蛋白的表达,逆转录聚合酶链反应(RT-PCR)检测PAR-1 mRNA的表达。【结果】脑脉Ⅱ号和脑血康均可显著性降低PAR阳性细胞数及PAR-1 mRNA吸光度比值(P<0.05或P<0.01),且两者作用无显著性差异(P>0.05)。【结论】脑脉Ⅱ号抑制脑出血后的PAR-1活化作用与脑血康相仿,这可能是其促进神经功能修复的主要机制之一。 [Objective] To study the influence of Naomai Ⅱ Capsule (NC- Ⅱ, mainly composed of artificial Calculus Bovis, Comu Bubali, Radix Gentianae, Rhizoma Polygoni Cuspidati, Hirudo, Herba Leonuri) on the expression of thrombin receptor protein, i.e., protease-activated receptors-1 (PAR-1), in rats with experimental cerebral hemorrhage. [ Methods ] One hundred and four rats were randomized into normal group ( N = 8), model group ( N = 32), NaoxueKang (NK) group (N=32) and NC-Ⅱ group (N=32), Except the normal group , the rats in other 3 groups were executed respectively 6 h, 24 h, 3 d and 7 d after cerebral hemorrhage, and protein expression of PAR-1 in cerebral tissue was examined by immunohistochemical method and PAR-1 mRNA expression by reverse transcription polymerase chain reaction (RT-PCR). [Results] NC-Ⅱ and NK markedly decreased the number of PAR-positive cells and optic absorption ratio of PAR-1 mRNA ( P 〈 0.05 or P 〈 0.01 as compared with those in the model group). The difference between the two groups was insignificant ( P 〉 0.05). [ Conclusion] NC-Ⅱ can inhibit the activation of PAR-1 after cerebral hemorrhage, the effect is similar to NK, and this probably one of its main mechanisms in promoting the repair of neurological function.
