摘要 目的研究白藜芦醇对大鼠心肌缺血再灌注损伤的保护作用,并进一步阐明其机制。方法建立心肌缺血再灌注损伤模型,持续观察标准Ⅱ导联心电图变化,测定乳酸脱氢酶(LDH),丙二醛(MDA)及超氧化物歧化酶(SOD)含量,确定心肌损伤程度。心肌细胞凋亡的检测采用逆转录聚合酶链反应检测bcl-2和bax mRNA的表达;Western blot检测bcl-2和bax蛋白表达。结果白藜芦醇能降低大鼠心肌冠脉结扎后ST段抬高,降低LDH和MDA水平,升高SOD水平,抑制bax而增强bcl-2的表达,减少心肌细胞凋亡。结论白藜芦醇对冠脉结扎诱发的大鼠心肌缺血再灌注损伤有保护作用,其机制可能与上调bcl-2蛋白表达,下调bax蛋白表达抑制心肌细胞凋亡有关。 Objective To investigate the protective effects of resveratrol against myocardiac ischemic/reperfusion injury and its mechanism. Methods Persistent observation of standard lead Ⅱ ECG in animal model of myocardiac ischemic/reperfusion injury. LDH, MDA and SOD were tested respectively to evaluate to degree of myocardiac damage. Apotosis of myocyte was tested by reverse PCR measuring mRNA expression of bcl-2 and bax gene. Protein expression of bcl-2 and bax was measured by Western blot. Results Resveratrol decreased the elevation of ST segment in rats after coronary ligation, lowered down levels of LDH and MDA, elevated SOD level, inhibited bax gene expression and up-regulated expression of bcl-2 gene, decreased apotosis of myocytes. Conclusions Resveratrol showed protective effects against myocytes ischemic/reperfusion injury induced by coronary ligation in rats, its mechanisms probably related to inhibition of myocytes apotosis through up-regulation of bcl-2 protein expression and down-regulation of bax protein expression.
