摘要
白血病的发生、发展与其相关基因的结构及功能异常密切相关,克隆白血病相关新基因有助于深入阐述白血病发生、发展的分子生物学机制,为白血病的诊断和治疗奠定基础。目前克隆新基因全长的方法主要有cDNA文库筛选法、cDNA末端快速扩增法、反向PCR和锅柄PCR、基于表达序列标签的全长cDNA电子克隆,用这些方法克隆的新的白血病相关新基因有ZFP91、HV126、白血病复发相关候选基因、急性白血病预后风险因子1、混合性白血病融合基因。
Generation and development of leukemia are closely associated with abnormality of structure and function of its related gene, cloning of novel leukemia related gene is helpful to elucidate molecular biological mechanisms in its genesis and development, and also provide a basis for its diagnosis and therapy. At present, there are several main methods used to clone full length of this new gene including cDNA library screening, rapid amplification of cDNA end,inverse PCR and panhandle PCR, silicon cloning based on expression sequence tag, through which the newly cloned gene related to leukemia are ZFP91, HV126, candidate gene related to leukemia relapse, d, outcome predictor in acute leukemia 1, myeloid/lymphoid or mixed-lineage leukemia fusion gene.
