多枝赖草、高羊茅的组织培养和愈伤组织诱变 被引量:4

摘要 以多枝赖草、高羊茅的种子为材料,在诱导培养基上进行暗培养,产生愈伤组织.愈伤组织经多次继代培养后仍可保持分化潜力.在分化培养基上进行光照培养后,可以分化出丛生状幼苗,并大量生根,形成再生植株.用EMS溶液对2种愈伤组织进行诱变处理,结果表明:不同体积分数的EMS对愈伤组织的致死率不同,随着体积分数的提高,致死率逐步升高.经统计,初步确定EMS溶液对多枝赖草、高羊茅愈伤组织的半致死体积分数分别为6 mL/L和8 mL/L. Tissue cuhure of Leymus multicaulis and tall rescue (Festuca arundinacea ) were optimized by using mature seeds as explants. The callus could keep the ability of differentiation after many times subcuhuring. On the medium of differentiation medium, many seedlings were induced from the callus, and grew out a lot of roots at the same time. Two kinds of callus were induced and handled by EMS (ethyl methane sulphonate). The results showed that the lethality rate of callus was increasing when we increased the concentration of EMS. By analyzing the results,the median lethal concentration of EMS for the callus of Leymus multicaulis and tall fescue was 6 mL/L and 8 mL/L respectively.
