摘要
目的研究转化生长因子β1(TGF-β1)在慢性移植物肾病(CAN)发生发展中的作用以及免疫抑制剂对TGF-β1的影响。方法采用实时荧光定量PCR检测15例CAN和22例非CAN患者外周血淋巴细胞TGF-β1 mRNA表达,分析CAN、免疫抑制剂和TGF-β1 mRNA表达3者之间的关系。结果CAN组TGF-β1 mRNA表达明显高于非CAN组,分别为0.1 55±0.027和0.123±0.028(P〈0.01)。服用环孢素(CsA)的患者TGF-β1 mRNA表达明显高于服用他克莫司(FK506)者,CAN组分别为0.170和0.137(P〈0.01),非CAN组分别为0.135和0.106(P〈0.01)。TGF-β1 mRNA表达与肾移植受者血肌酐(r2=0.77,P〈0.01)和血尿素氮(r2=0.63,P〈0.01)呈良好线性关系。结论CsA能引起肾移植受者外周血淋巴细胞TGF-β1 mRNA高表达,后者可诱发和促进CAN的发生与发展。
Objective To study the relation of TGF-β1 in the development and progression of chronic allograft nephropathy (CAN) and the effect of immunosuppressants on TGF-β1. Methods The expression of TGF-β1 mRNA in peripheral blood lymphocyte(PBL) was detected in 15 CAN pa dents and 22 non-CAN patients, who were divided into CsA or FK506 groups respectively. The relation of CAN development, TGF-β1 expression and immunosuppressant were observed. Results The expression of TGF-β1 mRNA was higher in CAN group than that in non CAN group significantly, 0.155±0.027 vs 0. 123±0. 028, P(0. 01. The expression of TGF-β1 mRNA was higher in CsA group than that in FK506 group, 0. 170 and 0. 137 in CAN group, and 0. 135 and 0. 106 in non-CAN group, both P〈0.01. The expression of TGF-β1 mRNA was negative correlated with allograft func tion, P〈0.01. Conclusions CsA is related to the higher expression of TGF-β1 mRNA, which may induce the development of CAN.
